$Honighäuschen (Bonn) - It is thirty years since the technique of high-yield preparation of isolated hepatocytes, by collagenase perfusion of the liver, was published. The original method described by Berry and Friend has undergone many minor modifications by other workers, and the two-step procedure introduced by Seglen in 1976 has become the most frequent way to prepare hepatocyte suspensions. An important development introduced by Bissell in 1973 was the use of the cells as the first step in monolayer culture. The availability of the isolated hepatocyte preparation as cells in suspension or culture has undoubtedly facilitated research on the liver. This was emphasised in our book, published (with Dr. Greg Barritt) in 1990, which described in detail methods of preparation and the properties of the isolated hepatocytes. It also discussed the usefulness of the preparation for the study of intermediary and xenobiotic metabolism, calcium ion transport, and the growth and differentiation of hepatocytes in culture. The book also touched briefly on a range of specialised techniques, including peri fusion, subcellular fractionation, transplantation, cryopreservation and measurement of intracellular pH. Although standard procedures for the manipulation of hepatocytes have not changed a great deal in ten years, they have undoubtedly been refined. This applies particularly to hepatocyte culture techniques, cryopreservation, and even to preparation of hepatocyte suspensions, where it is now feasible to use purified enzymes. There is also much more emphasis on the use and study of human hepatocytes, particularly in the field of pharmacology and therapeutics.$

Honighäuschen (Bonn) - This book came about as a result of a review I had written earlier on fea tures of cellular changes occurring during anuran metamorphosis. Only a limited treatment of this subject was possible in such a circumscribed work and only specific examples of organic change were dealt with. Thus the sins of omission weighed heavily, for so much information could not be included to provide a more comprehensive and authenticated account of the elaborate, complex, and far-reaching changes that an aquatic larva undergoes to become a terrestrial froglet. A good deal of my working life has been spent investigating amphib ians, especially their larval developmental morphology during metamor phosis, first at the level of light microscopy and in later years by electronmicroscopy. Initially I was particularly concerned with morpho logical homologies of a variety of larval structures, such as the cranial and pharyngeal skeleton and the nerves and musculature, in order to learn more about amphibian phylogeny, for during my pre-and early postgrad uate years G. R. Beer and D. M. S. Watson inspired an undying interest in and respect for vertebrate comparative anatomy. However, it now seems to be that amphibian phylogenetic relationships are best dealt with by the paleontologists, so ably demonstrated by D. M. S. Watson and A. S. Romer and the contemporary enthusiasts in this field like A. L. Panchen, R. L. Carroll, E. Jarvik, and K. S. Thompson among a host of others, particularly in the USA.

Honighäuschen (Bonn) - This volume focuses exclusively on those endocytic processes that sequester proteins by a selective, receptor-mediated mechanism. In such an endocytic process, cell surface receptors specifically bind protein ligands and localize them to specialized invaginations of the plasma membrane. These regions are coated pits, so named because they are lined on the cytoplasmic face with an ordered array of the protein, clathrin. It is this 'coat' which provides their characteristic electron microscopic image. Subsequently, these regions pinch off to form coated vesicles which rapidly lose their 'coat' and then fuse with other organelles or the plasma membrane. The hallmarks of ,this process are the specific receptors, coated pits, coated vesicles and an ordered sequence of transit events leading to delivery to selected locations. Receptor recognition, specific disposition of the endocytosed ligand and the existence of recep tor-ligand complexes at highest density in coated pits define the process as selective and concentrative. This topic has received ever increasing attention during the past few years. The evolving mechanisms are especially exciting because they come at a time when the conventional views based on thermodynamic arguments suggest that proteins should not be able to cross into the cell. Receptor-mediated endocytosis, however, reconciles the view that biological membranes should be impervious to macromolecules with the evidence that certain mac romolecules do gain entrance into the cell. During the last few years this field has been stimulated by studies on the uptake and processing of low density lipoproteins (LDL) by cells.
